M230I In pH1N1 In Southeast Wisconsin

Recently released sequences by JCVI at Genbank include A/Wisconsin/629-D00337/2009, which has HA M230I.  M230I is in all contemporary seasonal flu (H1N1, H3N2, influenza B) and previously was of concern because of its appearance in H5N1 isolates from the Gharbiya cluster and other fatal H5N1 infections in Egypt.  Although the 1918 H1N1 sequence has M230, the acquisition of M230I by all contemporary seasonal flu sequences suggests it offers a selective advantage in humans and its acquisition by swine H1N1 raises concerns that it is involved in adaptation to the current human target population.

The above isolate is from southeast Wisconsin and was collected December 15, 2009, so recent spread is unknown because there is only one 2010 HA sequence from Wisconsin.  A change at the adjacent position, N231K has been circulating in southeast Wisconsin in the fall of 2009, but N231K is not in the above isolate.  The acquisition of changes in or adjacent to the receptor binding domain is a concern because of potential effects on tissue tropism, as well as immune recognition.

Release of additional 2010 sequences from Wisconsin and the rest of the US would be useful.
